Experimental and computational methods for determination of local values of Young´s modulus in polycrystalline materials

The bachelor thesis combines computational and experimental methods for determination of local Young's modulus of polycrystalline materials. Quantum-mechanical calculations are used for theoretical determination of anisotropic elastic properties and the results are compared with those from nanoindetation measurements. The aim of the bachelor thesis is to apply these two methods to elemental iron as a pre-step for future research of binary Fe-Al alloys.
Determination of masonry modulus of elasticity in the direction of bed joints

The determination of strain properties of masonry in the direction parallel to bed joints is a fundamental pre-requisite for designing structures, where masonry is subjected to horizontal stresses (e. g. strengthening with prestressing). The diploma thesis summarized techniques of determination of masonry modulus of elasticity presented in available literature. The goal is to suggest suitable methodology of determination of masonry modulus in direction parallel to the bed joints. In the practical part of the diploma thesis is that methodology verified by experimental test and results of tests are analyzed and discussed.
The impact of freezing of arterial tissue on its stress-strain response

This bachelor thesis deals with freezing of the arterial tissue at -20 °C and its impact on mechanical properties of artery. The introductory part focuses on basic medical terms related to the issue followed by presentation of scientific publications dealing with artery tissue freezing. The attention is mainly paid to measurements and evaluation of results. The main part of the thesis contains description of the methodology of biaxial tensile test of porcine artery tissue with process of measuring the artery’s Young's modulus. Statistical analysis is used to determine the impact of freezing on mechanical properties of the artery. Description of limitations of this experiment is also included.
Stiffness anisotropy of Brno Tegel determined by continuous loading in triaxial tests

This Thesis deals with the determination of Young's modulus and Poisson's ratio from the measurement of small strains (10-3 - 10-5 ) in triaxial tests on the overconsolidated Miocene clay from Moravia (Brno Tegel). The tests were performed in a hydraulic triaxial chamber, fitted with submersible local LVDT sensors, which were fixed directly on the sample. The tests were carried out on undisturbed samples of standard height and diameter (76x38 mm). There were three types of tests - isotropic loading probe, axial loading probe and shear test to the failure. The measured data were analyzed and the results were discussed. The research part of the Thesis briefly describes previous works on stiffness of Brno Tegel and also some works on stiffness of similar material to Brno Tegel - London Clay.

Nuclear myosin 1 and its role in the regulation of plasma membrane tension

Myosin 1c (Myo1c) is a molecular motor involved in regulation of tension-gated ion channels, exocytosis, endocytosis, motility and other membrane-related events. Moreover, it acts as a dynamic linker between the cell membrane and cortical actin network, contributing to the maintenance of plasma membrane tension. In contrast, nuclear myosin 1 (NM1), an isoform of Myo1c, has been described only in the nucleus where it participates in various nuclear processes, including transcription or chromatin remodeling. However, although traditionally regarded as exclusively cytoplasmic or nuclear, all myosin 1c isoforms participate in nuclear functions and they are present in the cytoplasm as well. The main focus of this study was to characterize the functional significance of NM1 in the cytoplasm. We have found that NM1 localizes to plasma membrane and shows a uniform punctuated distribution with a high concentration at the cell periphery. Moreover, atomic force microscopy measurements of mouse NM1 KO fibroblasts revealed a significant increase in an overall plasma membrane elasticity in comparison to WT cells, indicating a disruption in the regulation of plasma membrane tension caused by the loss of NM1. Young's modulus of heat-resistant fibrous composites at elevated temperatures

Elastic behaviour of unidirectional composites with matrix derived from cured polysiloxanes and reinforced with SiC (Nicalon) or alumina (Nextel) fibres was measured at temperatures up to 1300řC in order to assess limits of their potential utilization as structural material in air environment.
